Abstract

The objectives of this project are: 1. To create an immunologically relevant genomic resources for the chicken (delivered by workpackage 1), made up of clusters of immunologically important genes, based on a resource of BAC end sequences (checked by milestone M1 and will give deliverables d1, d2) and delineating the repertoire of gene transcripts within these clusters (deliverable d3). 2. To construct a multi-tissue normalised cDNA library enriched in full-length clones (delivered by workpackage 2), representing the majority of chicken transcripts in immune tissues with active respones (deliverable d4). 3. To characterise a set of 30,000 cDNA clones from the normalised library by sequencing their ends, half of which should represent unique gene transcripts (workpackage 2, deliverable d5). To generate a specialised miniature cDNA array of non redundnat cDNA clones for up to 15,000 immune gene transcripts (delivered by workpackage 3, checked by milestone M3, M4) and use it to monitor the level of expression and tissue specificity of the corresponding genes (milestone M5, deliverables d6, d7). To identify candidate genes involved in resistance to Marek's disese, coccidiosis, and infectious bursal disease, which are among the principal pathogens of poultry in Europe, by exploring transcription profiles using well defined biological samples derived from resistant and susceptible birds (workpackage 4, deliverable d8, d9, d10) To establish genotyping and expression profile-based tests for field testing by the poultry industry (workpackage 5, deliverables d11, d12).
