Abstract
The objectives are to identify host pig macrophage proteins that bind to ASFV encoded proteins in order to predict functions for the virus encoded proteins. This will be achieved by screening a pig macrophage cDNA library with 7-10 different ASFV genes using the yeast two-hybrid screen. Sequences of inserts in clones encoding interacting proteins will be determined and the interactions confirmed by in vitro binding, co-precipitation from cell extracts and co-localisation in cells. Predicted functions for the virus encoded proteins will then be tested using methodology appropriate for that function.
