Abstract

Isolation of ribosome-associated mRNAs from foot-and-mouth disease virus infected cells should permit the identification of specific mRNAs which are translated when cap-dependent protein synthesis is inhibited. This will be achieved by hybridisation of labelled probes derived from these mRNAs to pre-existing cDNA libraries. Some mRNAs with short unstructured 5' NCRs may be translated with little requirement for intact eIF4F. However, other mRNAs translated within the infected cells may contain cellular internal ribosome entry site (IRES) elements. The properties of such cellular IRES elements will be characterised and compared to the well known picornavirus IRES elements.
