Abstract

The pathogenesis of disease following oral challenge of hamsters with the 263K scrapie strain is being investigated. The spread of TSE infection from sites of uptake in the gut is being traced using accumulation of disease associated forms of PrP as a surrogate marker for infectivity. The timing and sequence of PrP accumulation in the gut, lymphoid tissues, peripheral nervous system and central nervous system are being investigated using immunocytochemistry. This will indicate, not only which tissues become involved, but also the particular cells that are affected within these tissues. An important part of this project is to trace the neuroanatomical spread of infection from the periphery to the central nervous system.
