Abstract

Cellular prion protein (PrPC) is normally found attached to plasma membranes by a C-terminal glycolipid anchor. This glycoprotein is the precursor of PrPSc, a disease-specific isoform, which accumulates in brain and other tissues during the pathogenesis of scrapie, BSE and other transmissible spongiform encephalopathies. PrPSc differs in shape from PrPC : it is less soluble, more aggregated and more protease-resistant than its normal isoform. Some monoclonal antibody (mcAb) epitopes on the surface of PrPC are masked in PrPSc and the mcAb's to the codon 90-120 region of the molecule (such as 3F4) can only recognise PrPSc efficiently after it has been disaggregated and denatured. The IAH have developed an ultra-sensitive, time-resolved fluoroimmunoassay for PrP using two mcAb's, FH11 and 3F4. This assay is being used to measure PrP in brain extracts and body fluids.
