Abstract

The most highly purified infectious fractions from scrapie- infected tissues contain an abundance of polymeric PrPSc but also various amounts of non-PrP polypeptides, glycosaminoglycan lipid and nucleic acid fragments. Denaturation of these fractions leads to the depolymerisation of the aggregates and controlled renaturation allows the repolymerisation of these complexes without the loss of infectivity. Experiments will be carried out to determine whether the stoichiochemistry of the components alters during this cycle of denaturation and renaturation and whether the removal of particular components alters the structure of the infectious titre of the infectious fraction complex.
