Abstract

We have carried out genomic studies (cloning and sequencing the gene and its promoter, identifying promoter polymorphisms between inbred lines of chickens, and mapping the gene) on several chicken cytokine genes including interferon- (IFN- ), interleukin-2 (IL-2) and IL-8, in the process clarifying the identity of the latter two molecules. Both IL-2 and IL-8 show synteny of genetic location between chickens and mammals. We have expressed bioactive recombinant chicken IFN- and IL-2, and are raising monoclonal antibodies to them. We also cloned the genes for IFN- from several species of galliforms. The high degree of overall identity (87.8-97.6%) at the predicted primary amino acid sequence level of the protein, including the deduced IFN- receptor binding motifs, suggests that IFN- may be cross-reactive among these species.
