Abstract

Infection with the avirulent Portuguese non-haemadsorbing tick isolate Ourique/T88/3 of African swine fever virus was found to protect large white/Landrace crossbred pigs and inbred pigs of the dd haplotype from lethal challenge with the related, but virulent, Portuguese tick isolate Ourique/T88/1. When a sub-clinical systemic viraemia was detected, there was always an accompanying rise in the number of circulating CD8hi sub- population. Intra-venous inoculation of ascites fluid containing mouse IgG2a monoclonal antibodies 11/293 or 76/2/11 to total CD8+ porcine lymphocytes abrogated this protection with a correlated depletion of the CD8+ lymphocytes. Analysis of the circulating lymphocytes in the anti-CD8 treated pigs showed all the sub-populations of CD8+ lymphocytes were reduced. These results provide the first definitive evidence for the participation of cellular immunity in resistance to ASFV.
