Abstract

A method for accurately assessing numbers of cell surface expressed molecules (using quantitative flow cytometry) has been optimised, and used to examine expression of different cattle MHC class I genes. Consistent differences in the relative expression levels of class I genes have been observed, which are maintained during upregulation resulting from infection, or treatment with cytokines. Transcriptional control mechanisms are being investigated by sequencing the 5' upstream region of a number of class I genes. This study has utilised BAC clones (see 109/286) containing both classical and uncharacterised class I genes, and will lead to a greater understanding of their relative functional significance.
