Abstract
Progress in identifying cytokine genes in chickens has been slow because of limited DNA sequence homologies with mammals, lack of specific bioassays, lack of cross- reactivity, etc. The rapid systematic identification of these molecules would make a significant contribution to the development of chicken immunology in a wide context. In mammals, cytokine genes are often clustered on particular chromosomes. The immediate aims of this project are to map chicken genes known to be associated with these cytokine gene clusters in mammals. There are several examples of synteny between human, mouse and chicken chromosomes in terms of gene order/location, including the chromosomal location of IL-2 in all three species. The mapping information and the genes will then be used to clone YACs containing these genes and genomic sequences adjacent to them. An exon trapping system will then be utilised to generate a cDNA library containing representatives of all the genes encoded on the YACs. These will be sequenced to identify them where possible by sequence homology. Full length cytokine cDNAs will then be isolated.
