Abstract

The genetic map and yeast artificial chromosome (YAC) library of the chicken generated in related projects make it possible to compare the organisation of the chicken genome with that of other species. This project will investigate gene order and organisation in YAC contigs in an area of a microchromosome spanning the major histocompatibility complex (MHC) and also in an area of a large chromosome which contains a newly identified gene affecting resistance to Marek's disease. The work will complement related projects constructing a physical contig across the MHC region and identifying the Marek's disease resistance gene, while allowing detailed comparison of gene order with that in mammals for two representative areas of the genome. This work is part of the BBSRC PAGA programme with the objective of consolidating the UK base in comparative genome mapping, and will also contribute to the isolation and identification of genes involved in the disease process. The project will generate detailed comparisons of gene order and gene organisation for two regions of the chicken genome, and will provide the first estimates of the distances over which gene order is conserved between birds and mammals, and the extent to which this is disrupted by local duplications and micro- rearrangements. The project will also identify new genes within the chicken major histocompatibility complex (MHC) which may have influence on disease, and will contribute to the identification of the Marek's resistance gene on chromosome 1. Information regarding the degree of gene conservation between mammals and chickens will be used in related projects and by the chicken genome community generally. Individual genes isolated by this work, either in the MHC region or candidate genes for resistance to Marek's disease from chromosome 1 will be evaluated and exploited in collaboration with Ross Breeders (U K) Ltd.
