Abstract
Sequence determination of part of the genome of fowlpox virus revealed the presence of an open reading frame encoding a homologue of the mammalian plasma cell glycoprotein, PC-1, not found in any other viruses. PC-1 is known to have alkaline phosphodiesterase, pyrophosphatase but its biological function is unknown. The project will involve: (i) targeted deletion of FPV PC-1 to determine whether or not it is an essential function (if it proves to be non-essential the mutant virus will be examined to identify any phenotypic changes); (ii) analysis of transcription of FPV PC-1 RNA; (iii) expression of mammalian and FPV PC-1 in bacterial and poxviral expression vectors to allow: production of antisera (polyclonal and monoclonal) and biochemical analysis and (iv) use of anti-PC-1 sera to characterise native PC-1 and to localise the FPV protein within the infected cell. This project forms part of a larger programme to understand how interactions between fowlpox virus (FPV) and host cells at the molecular level influence virus host range and survival. FPV genome sequence determination revealed the an open reading frame encoding a homologue of the mammalian plasma cell glycoprotein, PC-1, not found in any other viruses (including the completely sequenced genomes of vaccinia and smallpox viruses).
