Abstract

As a contribution to an EU programme on the "Structure- function relationship of pectins and pectinolytic enzymes" we plan to determine the 3-dimensional structure and catalytic mechanism of pectin lyase A from A-niger. This enzyme is currently used in the pectin industry to process citrus pectin as a replacement for harsh inorganic acids. The programme will probe the catalytic mechanism of pectin lyase using site-directed mutagenesis and attempt to design new enzymes tailored to their industrial application. Enzyme structure-function relationships will also be examined using natural protein inhibitors found in plant species that apparently act to protect the plant from certain phytopathogenic microbes.
