Abstract
Colorectal cancer develops from adenomatous polyps that emerge gradually from aberrant fields of mucosal epithelial cells. Biomarkers are needed for research into the role of diet in the early stages of this common condition. We have developed 2 new methods to detect (Methylation-Specific PCR), and quantify (Methylation-specific restriction fragment length polymorphism) CpG island hyper-methylation in human faecal DNA, derived from colorectal epithelial cells. We now propose to use faecal samples to determine the occurrence of abnormal CpG island methylation, at 5 target genes, in both healthy volunteers and patients. We will also use a PCR-augmented genomic DNA subtraction technique to identify novel genes acquiring CpG island methylation early in the onset of neoplasia. Finally we will search for patterns of hyper-methylated genes that define early stages of the adenoma-carcinoma sequence, and design protocols for their use as faecal biomarkers in epidemiological and dietary studies.
