Abstract
Storage proteins are major determinants of the functional properties and end use properties of seeds, including the breadmaking quality of wheat. In order to understand these properties, and to exploit them in the food or other industries, it is necessary to understand how the proteins are synthesised, processed and trafficked in the cells of the developing seed. Work in this RO focuses on the basic biology of these processes, with emphasis on the gluten proteins of wheat. It includes studies of protein translocation into the lumen of the ER, folding and assembly in the ER lumen (including the roles of chaperones and PDI), trafficking through the secretory pathway (including the role of small GTP-binding proteins) and deposition into protein bodies. Objectives 1996 Continue studies of the role of small GTP- binding proteins in trafficking. Study the role of BiP and PDI in folding of gamma-gliadin in vivo and in vitro. 1997 Generate transgenic wheat expressing wildtype and trans- dominant forms of the tobacco Rab1 small GTP-binding protein under the control of a seed specific promoter. Examine developing grains from these lines for alteration in the deposition of protein bodies, including the composition by various different seed storage proteins (gliadins versus glutenins). Finish work on ectopic expression of the seed- specific protein oleosin.
