Abstract
Plants have versatile detoxification systems to counter the phytotoxicity of a wide variety of natural and synthetic chemicals (xenobiotics). An important cellular detoxification mechanism is the chemical modification of the xenobiotic by conjugation to endogenous metabolites such as glutathione or glucose, followed by the export of the conjugates from the cytosol to the vacuole or the apoplast. The aim of this project is to identify and characterise the key genes and proteins responsible for conjugation and transport in the cereals Hordeum vulgare (barley) and Triticum aestivum (wheat), and in herbicide-resistant populations of the grass weeds Alopercurus myosuroides (black-grass) and Avena spp. (wild oats). Special emphasis will be placed on ATP-binding cassette (ABC) transporters, in particular how their temporal and spatial expression affects the metabolism and fate of xenobiotics in planta. At the cellular level,conjugation and compartmentation mechanisms will be studied in cultured cells, protoplasts and isolated vacuoles using fluorescent and radioactive tracers. Expression analysis at gene and protein levels will be carried out by Northern blotting and immunolocalisation respectively.
