Abstract
The biosynthesis of the gibberellin (GA) hormones is regulated by intrinsic and environmental factors, many of which act on GA 20-oxidase (GA20ox), a rate-limiting enzyme that functions late in the pathway. The Arabidopsisis genome contains five putative GA20ox genes, some which have been shown to be expressed in a distinct tissue-specific manner. The hypothesis that these genes are involved in different developmental processes, such as stem extension, flower development and seed germination, will be tested by isolating lines with knock-out mutations in each GA20ox gene, either by screening populations of T-DNA insertion lines, or by specific gene silencing using RNA interference. Since there is evidence for partial redundancy for the GA20ox genes, multiple gene knock-outs will be produced, either by crossing the single gene mutants or knocking out several genes simultaneously by RNA interference. The phenotypes of the mutants will be correlated with their gene expression patterns, determined using reporter genes and by in situ hybridisation and immunolocalisation.
