Abstract
In vivo neurochemical sampling methods will be used to assess changes in basal and evoked-release of hippocampal, frontal cortex and striatal transmitters as a function of normal ageing in rats and mice and in transgenic mouse lines that express various aspects of brain pathology and cognitive dysfunction associated with Alzheimer¿s disease. Therapeutic actions of novel anti-cholinesterase inhibitors or drugs affecting other transmitter systems affected will be assessed both in terms of in vivo neurochemical changes and altered cognitive functioning. Overall therefore this project aims to identify genes and transmitters specifically involved in cognitive decline during ageing or neuropathology and is therefore directly relevant to Foresight H&LS priorities in molecular and cellular Neurobiology, cognitive sciences and ageing. The project involves extensive national and international, academic and industrial collaborations which will all aid discovery of novel drugs for treatment of Alzheimer¿s disease.
