Abstract
The binding site on E-cadherin for the integrinalpha-E beta-7 will be identified using domain deletion mutants and point mutations of E- cadherin. The functional significance of this receptor-ligand interaction for mucosal immunity and for the integrity of mucosal epithelia will be investigated by genetically modifying the activation state of the integrin in transgenic mice. The project is multidisciplinary in that it has depended on the availability of biophysical/structural data on E-cadherin (atomic coordinates) from our collaborator together with cellular, molecular and whole animal physiological studies in our laboratory. This work is directly relevant to the H&LS Foresight priority on Manipulating the immune system (control of chronic inflammation) because it addresses fundamental mechanisms in mucosal immunity that are relevant to the pathogenesis of inflammatory diseases of the gut. It will be exploited to improve understanding of these processes and may lead to new strategies for therapy. In addition, the work will produce new research reagents. We collaborate with the Ontario Cancer Institute and St Bartholomews Hospital to facilitate the research and keep us in touch with the clinical situation.
