Abstract
Classical MHC class I genes of Rattus norvegicus, the laboratory rat, will be cloned and expressed. An attempt will be made to obtain complete sequence data on the 13 alleles of the RT1.Alocus identified in laboratory rat stocks world-wide in order to understand the relationship of these alleles and to relate sequence to peptide binding characteristics. The rat is the most widely used model animal for transplantation experiments, and the outcome of this research is expected to be of primary interest to a wide number of researchers. One of the anticipated commercial benefits will be the assembling of a kit allowing the typing of rat strains and cell lines for their MHC. This integrative biological project involves molecular cloning, expression and characterisation at the biochemical and immunological levels. We collaborate mostly with German and Japanese groups for this work. This work is primarily relevant to H&LS Foresight Priority Immunomanipulation because understanding of antigen presentation is the key to control of tolerance in, for example, chronic inflammation. The rat is the most widely used model animal for transplantation experiments, and the outcome of this research is expected to be of primary interest to a wide number of researchers. One of the anticipated commercial benefits will be the assembling of a kit allowing the typing of rat strains and cell lines for their MHC. The rolling programme grant work in the Laboratory forms a platform from which we have been able to attract both medical charity and pharmaceutical company funding and(in the latter case) collaboration.
