Abstract
The binding of signalling molecules to their receptors is fundamental process for many cellular responses. The goal of this project is to develop and apply new caged neurotransmitters and second messengers that will permit 3-dimensionally defined photorelease of these signalling molecules at synapses and intracellular sites via 2-photon photolysis (2PP). Two-photon excitation has proved extremely valuable for cellular imaging and, in principle, should allow 2PP of caged effectors. Unfortunately, currently available caged compounds are relatively insensitive to 2PP. This project will develop and test new chromophores that are effective for 2PP (In collaboration with Professor Graham Ellis-Davies, Philadelphia). The unprecedented spatial and temporal control of effector molecule application provided by these compounds will be exploited to examine the physiological properties of neurotransmitter receptors at single synapses (in collaboration with Professor Haruo Kasai, Okazaki) and of second messengers in HeLa cells and cardiac myocytes.
