Abstract

Work from our laboratory has identified Vav-1 as a key regulator of signal transduction by specific receptors in lymphocytes. Vav-1 is a multidomain protein and the challenge is to understand the roles of individual domains of this protein in specific biological processes. This project will therefore investigate gene function by using homologous recombination to introduce a single point mutation into the Vav-1 gene of the mouse. This mutation will inactivate one domain while retaining the function of other domains. Phenotypic analysis will focus on the effects of the mutation on development and function of lymphocytes in the intact organism. The analysis of signalling events that are Vav-dependent (for example Ca2+ flux, actin cytoskeleton dynamics, activation of serine kinase pathways and so on) will provide mechanistic insight. The project will thus lead us to an understanding of how a point mutation in a specific gene can give rise to alterations in function at the level of the whole organism.
