Abstract

Gene targeting is established for the analysis of gene function and the generation of mouse models of human disease. Conditional gene targeting enables the manipulation of genes in a given tissue and at a given time. This relies on the availability of promoters to drive gene expression in the desired spatial temporal manner. There is currently a severe deficit of such promoters. This proposal outlines a novel transgenic strategy to harness promoter specificity by enhancer trapping from P1 artificial chromosome (PAC) transgenes. Proof of concept will be investigated by PAC mediated expression of reporter genes and Cre recombinase.
