Abstract
Imprinted genes are those genes that are only active when they come from either the father or the mother. The mechanism of imprinting involves attachment of chemical groups (methyl groups) to the DNA to indicate whether the DNA comes from mother or father originally. The regions in which imprinted genes are differentially methylated may indicate important control regions for gene activity. We are eliminating these regions by deletion from the chromosomes in the mouse. This will enable us to determine their function in regulating imprinted gene expression. The identification of silencers, boundary elements and antisense transcripts is of particular relevance.
