Abstract

Although the crypt/villus axis of the small intestine is well characterised in terms of its cell biology, the precise identity of the stem cell population still eludes both in vitro and in vivo analyses. Recently, several potential new markers have been proposed, the majority of which mark both the putative stem cell population as well as early dysplastic intestinal lesions. This link between the stem cell population and early dysplasia has been acutely shown by the phenotype of mice mutant in the Wnt pathway, and also by the expression profiles of a number of Lef/Tcf target genes. This proposal seeks to exploit transgenic mice mutant within these pathways and their targets as reagents to facilitate the development of stem cell and stem cell related primary cultures, and subsequently to characterise these cultures through both microarray and subcutaneous injection.
