Abstract

Post-translational modification of protein substrates by the ubiquitin-like proteins SUMO and NEDD8 is required for important cellular processes in eukaryotes, ranging from cell cycle progression to stress responses. SUMO and NEDD8 attachment alters the function of modified proteins, changing their enzymatic activity, sub-cellular localisation or incorporation into multi-protein complexes. To date however only a limited number of targets for modification have been identified and straightforward proteomic approaches have been confounded by the requirement to preserve the modified state by cell lysis under highly denaturing conditions. Recently however we have circumvented these problems and our objective in this proposal is to characterise the SUMO and NEDD8 sub-proteomes and determine how their composition changes as cells progress the cell cycle and respond to stress.
