Abstract

The overall goal is to understand how membrane-binding domains insert into bilayers and bind phospholipids to target proteins to subcellular organelles. Targets include the PX domain (aim 1) and the DIX domain (aim 2). Structures of the phospholipid-bound and micelle-inserted states will be determined by NMR spectroscopy. The depth and angles of bilayer insertion will be measured using paramagnetic spin labels. The binding specificities and affinities for lipids will be measured. Regulatory mechanisms including protein interactions will be investigated. Residues that contact ligands will be mutated to test effects on subcellular localisation and in vivo interactions of full-length proteins.
