Abstract
The aim is to advance the understanding of the recently discovered ovarian BMP system. Building on our substantial preliminary work, this will be achieved using ovarian immunohistochemistry (to deteriner the spatio-temporal pattern of expression of BMPs and BMP receptors during bovine folliculogenesis) together with functional experiments involving well-defined in vitro models (bovine theca and granulosa cells in primary culture). Functional studies will use different approaches (including RNAi, immunoneutralisation, icc, ELISA) to test several hypotheses concerning autocrine/paracrine actions of BMPs. Given the lack of data on ovarian BMP secretion, importantly, we will develop novel immunoassays for bovine BMP-4, -6 and-7 and use these to study factors regulating BMP secretion by theca and granulosa cells.
