Abstract

The recently-developed technique of recombinogenic engineering will be extended to allow insertion of reporter genes precisely at the end of the protein coding region of cloned C. elegans genes. As well as reporting accurately on the transcriptional activity of a genes promoter, information on subcellular localisation of the endogenous gene product should also be provided with the C-terminally tagged proteins. The feasibility of using this approach for efficient C. elegans genomic investigation will be tested with a sample of twenty, muscle-specific genes, this tissue specificity being selected because of the structural order of muscle cells.
