Abstract

We shall elucidate the mechanism and specificity of pectin methylesterase and pectate lyase 9A by determining the three-dimensional structures of numerous, inactive, mutants co-crystallized with substrate and by mechanism-targeted kinetic studies of native and mutant proteins. Alternative mechanisms suggested by the structural studies will be distinguished by the presence or absence of kinetic isotope effects and the number of in flight protons. Using this combined approach we shall identify the key enzyme/substrate interactions involved along the reaction coordinate and elucidate mechanism. This work will reveal the key interactions responsible for rate enhancement by esterases and anti-beta-eliminating lyases.
