Abstract

This collaborative programme between St Andrews and Cambridge will explore fluorometabolite production in Streptomyces cattleya at the genetic, enzymatic and structural level. The programme develops from the recent identification of the first native fluorination enzyme, its cloning and solution of an X-ray structure of the enzyme. In particular SDM and crystallisation studies will be carried out to probe the mechanism of biological fluorination. The programme will extend to full sequencing of the biosynthesis gene cluster and over-expression of the relevant enzymes for structural studies. Knock out mutants and cloning of the entire fluorometabolite pathway will be explored such that new fluorinated products can be prepared and enzymatic fluorination can be manipulated into other micro-organisms with the prospects of developing the first biotechnological methods for organo-fluorine compounds.
