Abstract

The JAK/STAT pathway plays important roles in development and disease and is conserved between phyla, but shows redundancy in vertebrates. Drosophila melanogaster is one of the model systems for genetically identifying genes involved in signalling pathways, due to its genetic tractability, sequenced genome, limited redundancy and short regeneration time. We plan to isolate new JAK/STAT components using a sensitised F1 genetic screen. Ectopic dominant negative domeless expression in the adult eye reduces eye size, conversely ectopic ligand increases eye size and ectopic dominant negative receptor in the wing give extra veins. We plan to isolate mutants that suppress or enhance the pathway using these phenotypes as indicators. The role of novel JAK/STAT signalling components in mammalian systems will be investigated using mouse cell culture models.
