Abstract

Two related approaches will be used to physically characterise the contractile ring. First, affinity purification of epitope-tagged components of the contractile ring and the subsequent identification of the associated proteins by SDS-PAGE and MALDI-TOF mass spectrometry. Secondly, we have observed the release of the intact contractile ring from cells arrested in late mitosis, based upon this observation, we will develop a purification scheme for the entire structure. Subsequently a combination of genetic, biochemical and cell biology techniques will be employed to confirm the presence and function of newly identified proteins within the actomyosin ring.
