Abstract

The proper control of DNA replication fork progression after DNA damage is critical to cell survival and genetic stability. In bacteria, homologous recombination proteins are employed to facilitate the replication of a damaged chromosome. However, little is known about how vertebrate cells control the progression of replication forks on damaged chromosomes. To address this question, we have recently developed novel in vitro and in vivo assays by which to measure the rate of progression of vertebrate replication forks in the presence and absence of DNA damage. We will now extend and exploit these assays to characterise at the genetic, biochemical and structural level how vertebrate cells control replication fork progression. (Joint with BBS/B/06091)
