Abstract

The roles of the 4 type I PI3-K in the PI3K-dependent processes that characterise B cell function are still largely unknown. We have shown P110 delta has an important role in B cell development and function, but have evidence of a role for additional, so far unidentified, p110 subunits. Here we propose to study the role of p110beta, gamma and delta in B cell survival mediated by BlyS, CD40 and CD19 pathways. We will establish 1) if transgenic restoration of Bcl2 expression rescues B cell survival in p110 delta mutants; 2) the requirement for p110 delta in survival versus development of MZ and Follicular B cells 3) the requirement for PI3K in activation of Btk and the processing and presentation of antigen. Defining specific roles for individual PI3-K genes in different biochemical and cellular pathways will have a major impact on our understanding of the molecular mechanisms of these processes.
