Abstract
We propose a stepwise approach to the development of a series of permanent hybrid synthetic/biological constructs for future use in corneal reconstruction. We will combine advances in polymer synthesis with recent breakthroughs in the transplantation of cultured corneal and mucosal epithelium to produce a series of biomimetic hydrogel implants with optimum biomechanical and biocompatability properties. These will be modified to facilitate epithelialisation and wound healing but to discourage biofouling and inflammatory reactions. These implants will be designed to either be inserted cell-free and be colonised in vivo by the patients own cells, or alternatively, where the ability of the patients own corneal cells to colonise implants is compromised, we will develop methodology for pre-loading of the implants with autologous mucosal cells.
