Abstract

An ever-growing majority of protein therapeutics are glycosylated proteins. However, all current methods used for their large-scale production create mixtures of different carbohydrate patterns in these proteins. There is a pressing need for methods that create pure carbohydrate forms to allow truly accurate assessment of the effects of protein structure on biological activity and to ensure that contamination by potentially toxic forms can be avoided. The enzymes that are the focus of this application are rare examples of powerful catalysts for the selective introduction of carbohydrates onto protein surfaces. They therefore represent powerful untapped synthetic tools for use in the construction of pure glycoprotein forms of high potential value. Based on initial promising indications of activity, this proposal will assess the breadth of utility of these enzymes when used in conjunction with IP-protected protein glycosylation methods with a view to full exemplification and exploitation.
