Abstract

The project will make an initial contribution to the International Genome Research On Wheat (IGROW) initiative by generating deletion mutants of the internationally used wheat variety Chinese Spring. We will assess systematically the level of X-Ray dose required to create specific sizes of deletions in hexaploid wheat. We will use a defined region for which we have PCR-based markers as the means to identify deletions and estimate their size. Six doses of X-Ray will be used to create the mutagenised populations. The average gene spacing in the wheat genome is approximately 1 gene every 40Kb. It is predicted that for every 250 genes (a 10Mb region) which are deleted in hexaploid wheat, one might give an observable phenotype. We aim to produce deletions which range from 40kb to 10Mb in size. Such deletions would then encompass between one and 250 genes. IGROW have also asked that any sequences of specific regions are made public to help evaluate the genome analysis approach. My group has produced a contig of a 1Mb region on wheat Chromosome 5B. We have completed a 2x coverage sequence of this region. The shotgun coverage of this region will be increased to over 6x which from experience provides a high likelihood of uncovering most of the gene content of the region. We will then deposit the annotated sequence and its comparison with the rice and Brachypodium sequences into the public domain.
