Abstract

We will construct a genome scale metabolic network (GSMN) for common central metabolic pathways in S. coelicolor and Streptomyces avermitilis, defining a minimum functional Streptomyces network, in line with GSMN studies in other species. This will then be extended to encompass S. clavuligerus-specific pathways and fed with experimental data that examines the effect of: growth-limiting substrate; growth rate in chemostat culture and growth rate profile in cyclic fed batch culture on metabolic flux partitioning within the GSMN. Having completed the optimisation of the use of S. coelicolor DNA microarrays for S. clavuligerus transcriptomic experiments, we will compare up- and down- regulation of gene expression with up- and down- regulation of metabolic flux in the GSMN. Thus, with a high degree of confidence, we will able to carry out supervised (discriminant) cluster analysis of the relationship between terminal biosynthetic flux to clavulanic acid and that of all other reactions in the GSMN. This will enable us to design process feeds in order to optimise the most significant reactions identified above.
