Abstract

Understanding the function of SNARE proteins in vesicle trafficking has emerged as a major issue in cell biology during the past few years. SNAREs drive membrane fusion events, but are generally recognised as targets for, rather than elements of signal cascades per se. Our recent discovery of a plant SNARE in ABA signalling points to roles in cellular stimulus-response coupling that lie outside the boundaries of functions commonly attributed to these proteins. We have been hunting for proteins that contribute to SNARE-mediated events in Arabidopsis and have discovered a new interaction between a previously uncharacterised SNARE and one of six endomembrane (vacuolar) two-pore channels. These channels are thought to be important in propagating the Ca2 plus signals that are central to many signalling processes in plants. We find that a major domain of the SNARE affects channel activity in the vacuole. This project will examine the biological consequences of SNARE interaction with the channel protein and its impact on Ca2 plus signal transmission in the living cell.
