Abstract

UV-B induces physiological responses in plants, but very little is known about the mechanisms of UV-B perception and signal transduction. Through isolation and characterisation of an Arabidopsis mutant we have shown that a protein called UVR8 is specifically involved in these processes. UVR8 is structurally very similar to a human protein, RCC1, the guanine nucleotide exchange factor (GEF) for the small GTP-binding protein, Ran. RCC1 binds to chromatin and functions in several processes involving Ran-GTP, including nucleocytoplasmic transport and progression of the cell cycle. However, RCC1 is not known to function in UV-B responses. The aim of this project is to start to understand how UVR8 functions in plant UV-B perception and/or signal transduction. We will (i) establish whether UVR8 has the known properties of RCC1 homologues (nuclear localisation, chromatin binding and GEF activity); (ii) determine which parts of the UVR8 protein are essential to its function by testing complementation of the urv8 mutant with modified UVR8 proteins; (iii) examine whether UVR8 binds to nucleosomes and to specific histones; (iv) establish whether UVR8 functions through association with other proteins in a chromatin complex.
