Abstract
The aim of this work is to obtain detailed molecular information on the mechanism of proton delivery in ascorbate peroxidase. The specific objectives are: (i) to dissect the detailed mechanism of proton delivery in ascorbate peroxidase: (ii) to elucidate the individual roles of key active residues and the heme propionate in the proton transfer process. To deliver our objectives, we propose an integrated experimental approach that uses the combined power of kinetic and protein engineering techniques, together with high-resolution protein crystallography. The programme has been designed to be relevant across the peroxidase family and will allow us to make important connections between different peroxidases in a way that has not been previously possible.
