Abstract

The aim of this work is to provide a unified view of substrate binding and electron delivery in the class I heme peroxidase enzymes that is relevant across the whole peroxidase family. To achieve this, we have set a number of objectives that will allow us to make important connections between different peroxidases and that will deliver a level of comparative molecular details that has not been previously possible. The experimental approach will involve redesigning the substrate binding specificity and examining the mechanism of electron delivery in cytochrome c peroxidase, which has been a benchmark for our understanding of heme enzyme catalysis. The combined power of kinetic, spectroscopic and redox techniques, together with protein engineering and X-ray crystallography, will be used.
