Abstract
There are three main objectives: 1) To establish 3-D mammary epithelial cell (MEC) culture systems for apoptosis and remodelling studies; 2) To compare LIF/Stat3/apoptosis signalling in MEC with LIF/Stat3/self-renewal signalling in ES cells in 2-D and 3-D cultures; and 3) To use genetically modified ES cells and hybrid ES cultures to determine additional signalling components that regulate apoptosis and differentiation. This will provide insights into the maintenance of totipotency within a differentiating environment.
