Abstract

This project is focussed on resistance of the yeast Saccharomyces cerevisiae to organic acid preservatives, a problem in large-scale food preservation. It will entail screening of the gene deletant strain collection to produce a complete catalogue of genes required for low pH growth, for resistance to acetate, and for resistance to the more lipophilic sorbate. Parallel DNA chip analysis will provide the global patterns of gene regulation by transcription factors (War1, Smp1, Yil100w) that influence sorbate resistance. Regulation of War1, the factor most important for sorbate resistance be investigated in molecular detail. Finally the mechanistic basis of effects of the tryptophan biosynthetic pathway and of the Azr1 plasma membrane transporter on weak acid resistance will be analysed.
