Abstract

To avoid cell death, it is vital that the expression of restriction-modification genes is subject to strict temporal control such that restriction activity is delayed with respect to methylation. This delay is accomplished by means of a regulator protein, the C-protein. We have recently purified the C-protein from the AhdI R-M system and obtained crystals that diffract to 2.2 angstrom resolution. We propose to solve the structure of the protein and relevant protein-DNA complexes by X-ray crystallography, to define the precise DNA binding sequence(s) recognised by the protein, and to establish and subsequently modify the protein-protein and protein-DNA interactions that underpin this intricate genetic switch.
