Abstract

We have recently developed the first synthetic transmembrane signalling system that can respond to a reagent on one side of a lipid membrane by releasing a different molecule on the other side without having to transport a chemical species across the bilayer. We will modify this vesicle based system so that it can respond to a non-covalent binding event with a target messenger molecule (initially DNA duplex formation). Concurrently, we will modify the response by exploiting enzymatic catalysis, so that the signal on the interior of a vesicle is amplified. Combining these features will yield vesicles that can respond to our choice of targets by providing an amplified optical signal or release of a chemical (unrelated to the target messenger properties). These will be the building blocks of future integrated multicomponent molecular machines.
