Abstract
We propose a novel combinatorial selective labelling method for obtaining partial backbone NMR assignments which will be tested and developed using the protein GFP. The method will ameliorate the assignment of proteins exhibiting complex NMR spectra. The method will be combined with spin label derived distance measurements to provide a tool for high throughput structure calculations, using the SH3 domain and protein G as model systems. Both methods will also be combined with segmental labelling. The assignment method provides a tool for the rapid identification of protein interaction sites. We will use these methods to characterise the structural and functional properties of members of the bactericidal/permeability increasing (BPI) protein superfamily.
