Abstract
By studying the actinorhodin synthase from Streptomyces coelicolor we aim to determine how Type II PKS components combine to form a functional complex. We will elucidate the overall 3D architecture of the minimal system, determine how the components recognise one another, and also identify how they bind and stabilise assembly intermediates. Structures of downstream proteins involved in reduction, cyclisation and aromatisation along with their interactions with the minimal PKS components will also be studied.
