Abstract

REF is a core component of the TREX and exon junction protein complexes. It associates with a number of different proteins in these complexes and RNA is essential for mRNA export in yeast. We have identified conditions suitable for determination of the structure of REF by NMR techniques. We will determine the complete structure of the REF + and - RNA using NMR. We will also define the residues involved in the interactions with proteins such as Tap, Y14/MAGOH, DDX39 and ORF57 using chemical shift mapping. These studies should lead to structural insights into how this protein can interact with so many partners and RNA through its binding domains and will provide the first structure of a complete mRNA export factor which binds RNA.
